Gel Running Station Solutions
NONE OF THESE SOLUTIONS NEED TO BE AUTOCLAVED OR
STERILE FILTERED.
10X Tris Buffer pH 8.9
To make 1L:
242.28g Tris; add enough MQ H2O to dissolve solid, pH with HCl to
8.9, then bring up to final volume of 1000mL.
1X Tris Buffer
Dilute 10X Tris Buffer (above). 100mL 10X Tris added to 900mL MQ
H20.
10X Glycine Buffer pH 8.3
To make 1L:
144g Glycine
30g Tris
100mL 10% SDS
Add enough MQ H2O to dissolve solids, pH with HCl to 8.3, then bring
up to final volume of 1000mL.
1X Glycine Buffer
Dilute 10X Glycine Buffer (above). 100mL 10X Glycine added to 900mL
MQ H20.
10X Tricine Buffer pH 8.25
To make 1L:
121.14g Tris
179.20g Tricine
100mL of 10% SDS
Add enough MQ H2O to dissolve solids, pH with HCl to 8.25, then
bring up to final volume of 1000mL.
1X Tricine Buffer
Dilute 10X Tricine Buffer (above). 100mL 10X Tricine added to 900mL
MQ H2O.
50X TAE Buffer pH 7.6-7.8
To make 1L:
242.28g Tris
100mL of 0.5M EDTA pH 8.0
57.1mL Glacial Acetic Acid
Add enough MQ H2O to dissolve solids, pH with HCl to 7.6-7.8, then
bring up to final volume of 1000mL.
1X TAE Buffer
Dilute 50X TAE Buffer (above). 20mL 50X TAE added to 980mL MQ H2O.
Coomassie Stain Stock
To make 400mL:
Dissolve 4g of solid coomassie in 400mL MQ H2O.
Coomassie Stain
To make 1 bottle:
250mL Methanol
50mL Acetic acid
75mL Coomassie Stain Stock (above)
37.5mL MQ H2O
Combine in a dark colored bottle.
Destain Solution
To make 1L:
300mL Methanol
100mL Acetic acid
600mL MQ H2O
Combine and mix thoroughly. Pour into bottles at gel station.
